The longevity stack combines the two most fundamental cellular defense compounds: NAD+ (the energy and repair coenzyme) and glutathione (the master antioxidant). Both decline significantly with age, and their depletion is mechanistically linked to hallmarks of aging including mitochondrial dysfunction, genomic instability, cellular senescence, and chronic inflammation. NAD+ and glutathione form an interdependent cellular defense network. NAD+ drives mitochondrial energy production and activates sirtuins that regulate DNA repair. Critically, NAD+ is required to regenerate NADPH, which is in turn required to reduce oxidized glutathione (GSSG) back to its active form (GSH). This means NAD+ depletion directly impairs glutathione recycling, creating a cascade of compounding oxidative damage. Restoring both compounds simultaneously breaks this vicious cycle.
NAD+ and glutathione are biochemically interdependent. NAD+ is needed to generate NADPH, which is required to recycle oxidized glutathione back to its active form. Without adequate NAD+, glutathione supplementation alone has diminished effectiveness.
Research endpoints include oxidative stress markers (F2-isoprostanes, 8-OHdG, MDA), mitochondrial function, inflammation markers (IL-6, TNF-alpha, hsCRP), telomere length, NAD+ metabolome profiling, glutathione redox ratio, and epigenetic age clocks.
GlyNAC (glycine + N-acetylcysteine) supplementation studies by Kumar et al. demonstrated that restoring glutathione synthesis in older adults corrected oxidative stress, mitochondrial dysfunction, inflammation, and cognitive decline.
The GlyNAC study showed significant improvements at 16 weeks. NAD+ supplementation studies typically show biomarker changes from 4-8 weeks. The 16-24 week duration allows both compounds adequate time to show measurable shifts in aging endpoints.
